Bio-MEMS Seminar: "High-Throughput Analysis of Protein-Protein Interactions Using Droplet-based Microfluidics"

Abstract: In recent years droplet-based (or segmented-flow) microfluidic systems have emerged as a powerful technological platform for performing high-throughput chemical and biological experimentation. Since droplets may be generated at kHz frequencies, enormous experimental data sets can also be amassed in short times allowing complex statistical analyses of multi-component systems. Whilst droplet-based microfluidic systems are efficient in generating vast amounts of chemical and biological information, the development of sensitive detection methods is critical in allowing this information to be extracted and utilized. To this end, a variety of optical detection methods for probing segmented flows in real time have been developed. I will present the use of fluorescence resonance energy transfer (FRET) and fluorescence polarization (FP) detections in a droplet-based microfluidic system. Specifically, I will present protein-protein interactions between angiogenin, a small polypeptide implicated in angiogenesis and in tumor growth, and an anti-angiogenin antibody in a pico-liter droplet by using FRET and FP detections
