ABSTRACT: Campylobacter jejuni, enterotoxigenic Escherichia coli and Shigellasp. are major causes of bacterial diarrhea worldwide. After fine structural analysis of cell-wall polysaccharide structures, we explored the feasibility of developing a multi-pathogen conjugate vaccine using Escherichia coli proteins as conjugating partners for Campylobacter jejuniand Shigellapolysaccharides. We synthesized glycoconjugate vaccines in which Campylobacter jejunicapsular polysaccharides and Shigella flexnerilipopolysaccharides were conjugated to Escherichia coli adhesins. The vaccines were immunogenic in mice as monovalent, bivalent and trivalent formulations and raised functional antibodies capable of inducing hemagglutination inhibition. The World Health Organization has recently highlighted these conjugate vaccines has a preferred product to combat diarrhea in the developing world. The topics of conversation will focus on our approach to polysaccharide structural and conjugation chemistries taken along the path to human trials.
