Physical Biochemistry

The principles of physical chemistry are applied to the study of biological macromolecules. The course comprises transport properties, including the study of biomacromolecules by the techniques of diffusion, ultracentrifugation, and electrophoresis; enzyme kinetics, including the Michaelis-Menten equation and statistically valid methods for determining Km and V, competitive, uncompetitive and mixed enzyme inhibition and determination of Ki values; the temperature-jump method and a brief survey of other rapid reaction techniques, temperature effects and the application of H and S to the study of enzymes; spectroscopy, including infrared (IR) (rotational/vibrational) spectra, Ultraviolet-Visible Spectrophotometry (UV-VIS) and applications of fluorescence spectroscopy to biochemical problems.
